GE AKTAxpress Liquid Chromatography System
The AKTAxpress is a chromatography system designed for automated, multistep purification of affinity-tagged proteins and antibodies. The AKTAxpress gives you the excellent purity needed for structural and functional studies. Optimized protocols with a choice of up to four purification steps minimizes the need for chromatography expertise. Tag removal and maintenance procedures can be integrated into the purification protocols, eliminating manual interference during a run. Purification schemes for double affinity-tagged proteins are also supported.
AKTAxpress has a modular design that allows for a high degree of flexibility. A Single or a Twin pack can be upgraded with separate modules and up to 12 modules can be controlled from one computer. Each module is run independently and each may run different protocols even though control is through a single computer. On each module, up to four different proteins are purified serially meaning that if 12 modules are run in parallel, up to 48 different samples can be purified in one run. Algorithms in the system’s software allow the detection and storage of intermediate peaks in capillary loops, allowing the system to wash, equilibrate, elute, or re-inject samples on the different columns connected to the system.
With ÄKTAxpress, it is possible to create a range of automated single- and multistep protocols to provide high purity. The four techniques, affinity chromatography (AC), desalting (DS), ion exchange chromatography (IEX), and gel filtration (GF) can be combined in up to four steps (Fig 2). Double tagged proteins can be purified with protocol including two AC steps.
The system components include a pump with a flow range from 0.1 to 65mL per minute; an air sensor; conductivity meter with a dynamic range from 1uS/cm to 999.9 mS/cm an independent, static mixer with a volume of 0.37mL; a fixed-wavelength UV monitor; five 10mL capillary loops; five valves that control the movement of fluid in the system; and an integrated fraction collector that can hold 96-well plates or 24-well plates.
